High-Throughput QC
It is important to analyze compound collections from time-to-time to ensure that only high quality compounds are used in biological screenings. At Rilas we offer high-throughput LCMS QC of compound collections up to 3000 compounds/ week/instrument using our optimized UPLC/MS method with bespoke automated data analysis. Our LCMS QC provides:
- UV area% purity at 215 nm or any customer requested wavelength
- Confirmation of exact mass
High-Throughput Physico-Chemical Measurements
Kinetic Solubility: Measure UV or MS peak area in aqueous buffers including simulated gastric solutions relative to the peak area in DMSO or methanol solution
Stability: Samples in aqueous solutions including simulated gastric solutions analyzed over time and compare to the original purity
Glutathione Reactivity Measurement: This measurement assess the reactivity of covalent binders to free thiols in aqueous environment. Using high access of glutathione t1/2 can be calculated using first order reaction kinetics.
LogD at pH 7.4: Calculated based on retention time using a calibration curve
generated by a shallow water/acetonitrile gradient
Biomimetic Chromatography: Using HPLC methods to measure lipophilicity, phospholipid and protein binding and acid/base characteristics of biologically active molecules to predict a series of ADME Tox properties
